Design, Development and Characterization of Hyaluronic Acid Based pH Sensitive Liposomal In Situ Gel for the Treatment of Keratoconjunctivitis Sicca

The recent advances in ocular drug delivery have been successful surpassing conventional therapy, owing to its considerable limitations. This study aims develop and optimize in-situ liposomal system design once-daily liquid preparation treat Keratoconjunctivitis sicca. Liposomes of Hyaluronic acid were prepared using soya lecithin by a thin film hydration method. Eight trial formulations made keeping as fixed ratio varying the concentration cholesterol lecithin. optimized formulation was selected for hydrogel. Optimization carried out Central Composite Design (CCD) studying factors, amount effects on responses, % In- vitro release (R1) Percent Entrapment Efficiency (% EE) (R2). best predicted model R1 Quadratic and, R2 Two-Factor Interaction (2FI) without any significant lack fit. Optimum found be at 1:1:4 (drug: cholesterol: lecithin) showed sustained 31.28% 8 hours with highest %EE 47.36%. Liposomal gel retained better stability throughout period when stored refrigerator temperature displayed prolonged action (76.44%) compared (68.72%).
